Effects of pallidal neurotensin on haloperidol-induced parkinsonian catalepsy: behavioral and electrophysiological studies

Objective The globus pallidus plays a critical role in movement regulation. Previous studies have indicated that the globus pallidus receives neurotensinergic innervation from the striatum, and systemic administration of a neurotensin analog could produce antiparkinsonian effects. The present study aimed to investigate the effects of pallidal neurotensin on haloperidol-induced parkinsonian symptoms. Methods Behavioral experiments and electrophysiological recordings were performed in the present study. Results Bilateral infusions of neurotensin into the globus pallidus reversed haloperidolinduced parkinsonian catalepsy in rats. Electrophysiological recordings showed that microinjection of neurotensin induced excitation of pallidal neurons in the presence of systemic haloperidol administration. The neurotensin type-1 receptor antagonist SR48692 blocked both the behavioral and the electrophysiological effects induced by neurotensin. Conclusion Activation of pallidal neurotensin receptors may be involved in neurotensin-induced antiparkinsonian effects.

苍白球P物质对帕金森病僵直模型大鼠的影响

目的观察苍白球P物质对氟哌啶醇帕金森病僵直模型大鼠的影响。方法行为学实验采用大鼠腹腔注射氟哌啶醇诱发帕金森病僵直模型,苍白球分别微量注射生理盐水、NK-1受体激动剂SMSP、NK-1受体阻断剂SR140333B、SR140333B和SMSP的混合物,观察其对大鼠爬杆行为的影响。电生理学实验利用细胞外微压力注射上述药物,观察对苍白球神经元放电的影响。结果大鼠腹腔注射氟哌啶醇可立刻产生僵直症状,平均僵直潜伏期最大值出现在第30分钟,为(259.8±34.8)s。最小值出现在第50分钟,为(145.2±54.8)s。双侧苍白球微量注射SMSP可明显改善僵直症状,第10分钟平均僵直潜伏期为(10.4±3.4)S,第60分钟为(58.4±38.8)s,差异有统计学意义(P〈0.01)。双侧苍白球同时注入SR140333B和SMSP混合物可明湿阻断SMSP的抗僵直症状,第10分钟为(176.4±64.4)s,第60分钟为(139.2±59.7)s,与单纯注射SMSP相比,差异有统计学意义(P〈0.01)。进-步细胞外电生理记录结果显示SMSP可使氟哌啶醇大鼠苍白球神经元放电频率由(13.4±4.2)Hz增加至(17.5±5.6)Hz,平均升高(29.4±8.6)%(P〈0.05,n=13),该效应可被SR140333B完全阻断(SR140333B:(10.3±2.5)Hz,SR140333B+SMSP:(11.3±3.0)Hz,P〉0.05,n=8)。结论苍白球P物质通过增加神经元放电,改善氟哌啶醇所致的神经元兴奋性降低,从而解除基底神经节对丘腑及大脑皮层的抑制效应,提示其可能起抗帕金森病的作用。

5-HT对帕金森病模型大鼠丘脑底核神经元放电影响被引量:6

目的探讨5-羟色胺(5-HT)对帕金森病(PD)模型大鼠丘脑底核(STN)神经元放电的影响。方法大鼠右侧内侧前脑束注射6-羟基多巴胺(6-OHDA)制备PD鼠模型,2周后采用细胞外电生理记录法,通过玻璃微电极向STN微量加压注射5-HT(0.1 mmol/L),观察其对正常大鼠和PD模型大鼠STN神经元放电影响。结果 PD模型大鼠STN神经元基础放电频率和放电模式与正常大鼠相比差异无显著性。正常大鼠细胞外微量加压注射5-HT可使22个STN神经元中的7个放电频率增加(t=2.45,P<0.01),而使3个STN神经元放电频率降低(t=4.30,P<0.05)。在PD模型大鼠损毁侧,5-HT可使10个STN神经元中的5个放电频率增加(t=2.78,P<0.05),而使1个STN神经元放电频率降低。结论 5-HT可以调节正常及PD模型大鼠STN神经元电活动。
