大麦黄矮病毒运动蛋白在烟草中的瞬时表达被引量:2

[目的]快速鉴定运动蛋白基因在烟草中的瞬时表达,进一步研究该外源基因的功能。[方法]将大麦黄矮病毒的运动蛋白基因定向克隆到马铃薯X病毒载体上,得到重组的马铃薯X病毒,电击转化农杆菌后,利用农杆菌渗透注射技术注射到本生烟草的叶片中,观察病毒对烟草的侵染状况。[结果]渗透注射后第7天观察,重组病毒载体侵染的烟草系统叶有病毒侵染症状,而对照组未有此现象。对2组试验进行逐日跟踪观察,重组病毒载体侵染的烟草有较严重的病毒侵染症状和叶片卷曲现象,后期引起注射叶及系统叶坏死。而空病毒载体侵染的烟草只有轻微的病毒侵染症状,并能够恢复健康。对侵染的烟草进行RT-PCR检测,结果表明外源基因BYDV-MP在烟草体内进行了正常的转录和表达。[结论]BYDV-MP蛋白促进了PVX的系统侵染速度,加重了系统侵染的病毒症状,是病毒病症的决定因子;利用PVX表达载体表达异源MP的方法是可行的。

Biological Characteristics of Lepus timibus in Captivity

The biological characteristics of Lepus timibus in captivity, such as living habit, feeding habit, nutrition composition of main food, growth and development, molting rule, were studied systematically. The results showed that there were more than 30 kinds of common plants favored by L.timibus. The linear regression equation of weight gain was y=-23.223+30.929 x. The molting sequence in spring successively was head, dorsum, ear,both sides of the chest, abdomen, limbs, tails.

大麦黄矮病毒运动蛋白是RNA沉默抑制因子被引量:2

大麦黄矮病毒(barley yellow dwarf virus, BYDV)属黄症病毒科家族, 其基因组包含6个开放阅读框(open readingframes, ORFs)。将BYDV的6个基因分别克隆到pWEIMING101载体上, 得到重组基因。电击转化农杆菌后, 利用农杆菌瞬时表达方法渗透注射转GFP基因的本氏烟草16c植株的叶片, 在长波长紫外灯下观察GFP的表达, 并通过Northern blot证明所得现象。研究结果表明, BYDV的PAV株系ORF4编码的运动蛋白(movement protein, MP)是RNA沉默抑制因子, 其表达可以抑制局部和系统RNA沉默。BYDV-MP与GFP的双链RNA(dsGFP)共表达后仍能抑制RNA沉默, 荧光强度与叶片中GFP的mRNA和其沉默降解形成的siRNA的量有对应关系, 其N端核定位序列对抑制局部基因沉默起主要作用, 第5、6位氨基酸是抑制基因沉默的关键氨基酸。BYDV-MP单独渗透注射的部位均产生细胞死亡。

Functional Analysis of Homodimer Formation between Movement Proteins in Barley Yellow Dwarf Virus

[Objective] The aim was to investigate the dimer formation between the movement proteins(MP)in barely yellow dwarf virus by using the technology of bimolecular fluorescence complementation technology and to further study the relationship between MP homodimerization and viral movement.[Method] The DNA sequence of bimolecular fluorescent complementary vector containing cloning multiple cloning sites,35S promoter and terminator was cloned into the expression vector pCAMBIA1300,which replicates at a higher copy number in E.coli.Then,the BYDV-MP gene fragment was amplified in the presence of the whole BYDV-PAV cDNA sequence as template and the primers designed according to the BYDV-MP gene sequence from GenBank,cloned into the modified bimolecular fluorescent complementary vectors pCAMBIA1300-NE and pCAMBIA1300-CE.The resulting vectors were transformed into Agrobacterium by electroporation method and infiltrated into the tobacco leaf.Protein interactions were observed under fluorescence microscope.[Result] Yellow fluorescence could be viewed in the leaves co-infiltrated with Agrobacterium carrying pCAMBIA1300NE-MP and pCAMBIA1300CE-MP at 2-5 d post-infiltration,while yellow fluorescence could not be observed in negative control groups.[Conclusion] BYDV-MP formed homodimers in plant cells.The results can provide theoretical basis for further in-depth research about the movement process and mechanism of BYDV.

病毒基因沉默抑制因子与AGO蛋白结合抑制RNA沉默的研究进展被引量:1

总结了RNA沉默及病毒RNA沉默抑制因子(VSRs)的研究进展,以及病毒基因沉默抑制因子利用甘氨酸/色氨酸(GW)模型作为ARGONAUTE(AGO)钩和寄主发生RNA沉默的重要作用部位相互结合从而抑制基因沉默的研究机制。通过研究发现,在酵母、动植物细胞中,一些包含GW模型的蛋白质被认为是RNA沉默效应复合物中AGOs的重要协助者。结果说明,GW模型是一种能用于调节RNA沉默途径活性的万能的、有效的工具,用GW模型竞争性结合AGOs并抑制寄主基因沉默可能是一种被许多病原菌用于抵抗寄主RNA沉默的方法。

Synthesis of the Intermediate for Prothioconazole被引量:1

[Objective] The paper was to study the synthesis technology of the intermediate for prothioconazole: 1-chloro-1-chloroacetyl-cyclopropane. [Method] With 2-acetylbutyrolactone as the raw material, 3,5-dichloro-2-pentanone was synthesized by chlorination and hydrolysis, and1-chloro-1-chloroacetyl-cyclopropane was synthesized by further cyclization and chlorination. [Result] The optimized synthetic process was determined by optimizing reaction conditions of each step. The overall yield of four-step reaction was 76.7%, and the product was verified by1 H NMR and13 C NMR. [Conclusion] The chlorinated product of 2-acetylbutyrolactone was directly hydrolyzed in acid solution without separation, and the final product was obtained by chlorination of cyclization product in the presence of special catalyst methanol. The whole process was smooth and the yield of the reaction was high, with high value for industrial application.

Cloning and Expression of Wheat Heat-shock Protein 60 (HSP60) Gene in E.coli被引量:2

[Objective]The aim was to construct the wheat heat-shock protein 60 (HSP60) gene in prokaryotic expression vector and express HSP60 efficiently in E.coli. [Method]According to the wheat HSP60 gene sequence in GenBank,a pair of primers P1/P2 were designed and synthesized. The wheat HSP60 gene fragment was amplified from the wheat RNA by RT-PCR and inserted into bacterial expression vector of pGEX-4T-1. The construct of pGEX-4T-1-HSP60 was subsequently transformed into E.coli BL21. [Result]The construct of pGEX-4T-1-HSP60 was verified by restriction endonuclease digestion and sequenced. Compared with the sequences of wheat HSP60 genes in GenBank,homology accounted to 100%. Expression of the GST-HSP60 fusion protein was induced with IPTG. Its molecular weight was about 90 kD. The result was identified by electrophoresis of SDS-PAGE. Expression of the protein bands was consistent with the expected size. [Conclusion]The recombinant prokaryotic expression vector in pGEX-4T-1-HSP60 was constructed successfully and expressed stably in E.coli BL21. This will lay the foundation for further study on the functions of the protein and its mechanism.

植物抗病毒机制是目前研究的热点.在长期的进化过程中,植物形成了一系列复杂有效的防御机制来抵御、破坏病原物的侵染.近年来,基因沉默作为一个重要的细胞内防御外源核酸的机制,越来越受到科学家的重视.对这一领域的研究不仅能够了解基因沉默的机理,并且可以为利用该机制构建抗病毒转基因植物提供理论指导和技术支持.

应用马铃薯X病毒(PVX)载体研究大麦黄矮病毒运动蛋白(BYDV-MP)核定位信号对PVX病毒运动的影响。我们将BYDV-MP克隆到PVX改造载体pGR107中,同时用GFP作为指示蛋白,研究BYDV-MP对异源病毒PVX系统运动的影响。侵染烟草发现BYDV-MP能够在PVX载体中表达并能加强病毒的系统侵染;将PVX编码系统运动蛋白25kD基因进行缺失突变,重复上述试验发现BYDV-MP能够补偿PVX系统运动;将BYDV-MP的N端的第五、六位氨基酸和第七位氨基酸进行替换突变,侵染烟草发现BYDV-MP的N端的第五、六位氨基酸突变不能完全抑制PVX系统运动,但是可以延迟并减弱PVX系统运动;BYDV-MP的N端的第七位氨基酸突变能够完全抑制PVX系统运动。

The epidemiology of highly pathogenic porcine reproductive and respiratory syndrome( HP-PRRS) in Changping District,Beijing City was investigated from 2010 to 2015. Through statistics of vaccine,immune procedure and antibody levels,the epidemic prevention effect of HP-PRRS in Changping District was scientifically assessed.
