This paper reports that an optical diagnostic system consisting of Mach-Zehnder interferometer with a phase shift device and image processor has been used for study of the kinetics of protein crystal growing process. The crystallization process of protein crystal by vapour diffusion is investigated. The interference fringes are observed in real time. The present experiment demonstrates that the diffusion and the sedimentation influence the crystallization of protein crystal which grows in solution, and the concentration capillary convection associated with surface tension occurs at the vicinity of free surface of the protein mother liquor, and directly affects on the outcome of protein crystallization. So far the detailed analysis and the important role of the fluid phenomena in protein crystallization have been discussed a little in both space- and ground-based crystal growth experiments. It is also found that these fluid phenomena affect the outcome of protein crystallization, regular growth, and crystal quality. This may explain the fact that many results of space-based investigation do not show overall improvement.
在地面实验中,使用粒子图像测速测温(Particle Image Velocimetry and Thermometry,PIVT)技术测量了单滴热毛细迁移时液滴周围同时刻的速度场和温度场。选用密度相近的豆油和硅油作为实验系统的母液和液滴。实验结果表明单滴周围的温度场扰动不同于线性理论预测的随到液滴距离的平方成反比,而是距离一次方成反比,这是由于重力引起的浮力效应造成的,地面实验中重力作用不能忽略。
Results from a space experiment on bubble thermocapillary migration conducted on board the Chinese 22nd recoverable satellite were presented. Considering the temperature field in the cell was disturbed by the accumulated bubbles, the temperature gradient was corrected firstly with the help of the temperature measurement data at six points and numerical simulation. Marangoni number (Ma) of single bubble migrating in the space experiment ranged from 98.04 to 9288, exceeding that in the previous experiment data. The experiment data including the track and the velocity of two bubble thermocapillary migration showed that a smaller bubble would move slower as it was passed by a larger one, and the smaller one would even rest in a short time when the size ratio was large enough.
Liquid mixtures of water and deuterium oxide as the liquid phase, were used to match the density of charged colloidal particles. Kossel diffraction method was used to detect the crystal structures. The experiments under the density-matched (g=0) and unmatched (g=1) conditions are compared to examine the influence of gravity on the crystal structures formed by self-assembly of 110 nm (in diameter) polystyrene microspheres. The result shows that the gravity tends to make the lattice constants of colloidal crystals smaller at lower positions, which indicates that the effect of gravity should be taken into account in the study of the colloidal crystals.
