Inhibition of proliferation,migration,and invasiveness of bladder cancer cells through SAPCD2 knockdown

Introduction:Bladder cancer(BC)has a high incidence and mortality rate worldwide.Suppressor anaphasepromoting complex domain containing 2(SAPCDC2)is over-expressed in a variety of tumors.Objectives:This study investigated the effects of SAPCD2 knockdown on BC cells.Methods:T24 and UMUC3 cell models and the xenografted BC tumor model with SAPCD2 knockdown were established to observe the malignant phenotype of BC cells by cell counting kit-8 assay,colony formation test,wound healing,and Transwell assay,mRNA and proteins expressions were measured with quantitative real-time polymerase chain reaction,western blotting,and tissue immunohistochemistry.Lithium chloride agonist on the Wnt/β-catenin pathway was used to clarify the molecular mechanism of SAPCD2 knockdown.Results:SAPCD2 expression was significantly higher in BC cell lines than in SVHUC-1 cells.SAPCD2 knockdown inhibited viability and cloning,hindered the G0/G1 phase of the cell cycle in UMUC3 and T24 cells,and decreased the migration and invasiveness of BC cells.SAPCD2 knockdown inhibited expression levels of cyclin D1,cyclin B1,N-cadherin,vimentin,Snail,β-catenin,c-Myc,and cyclin-dependent kinase 4,while the P21 and E-cadherin were raised by SAPCD2 knockdown.Furthermore,lithium chloride reversed the effects of SAPCD2 knockdown on the expression levels of the above proteins in UMUC3 and T24 cells.In vivo,SAPCD2 knockdown inhibited the volume,weight,and expression of Ki-67 andβ-catenin in tumors and increased the E-cadherin expression.Conclusion:SAPCD2 knockdown inhibits the malignant phenotype of BC via a pathway involvingβ-catenin.

Knockdown of HE4 suppresses tumor growth and invasiveness in lung adenocarcinoma through regulation of EGFR signaling

It has been shown that the high expression of human epididymis protein 4(HE4)in most lung cancers is related to the poor prognosis of patients,but the mechanism of pathological transformation of HE4 in lung cancer is still unclear.The current study is expected to clarify the function and mechanism of HE4 in the occurrence and metastasis of lung adenocarcinoma(LUAD).Immunoblotting evaluated HE4 expression in lung cancer cell lines and biopsies,and through analysis of The Cancer Genome Atlas(TCGA)dataset.Frequent HE4 overexpression was demonstrated in LUAD,but not in lung squamous cell carcinoma(LUSC),indicating that HE4 can serve as a biomarker to distinguish between LUAD and LUSC.HE4 knockdown significantly inhibited cell growth,colony formation,wound healing,and invasion,and blocked the G1-phase of the cell cycle in LUAD cell lines through inactivation of the EGFR signaling downstream including PI3K/AKT/mTOR and RAF/MAPK pathways.The first-line EGFR inhibitor gefitinib and HE4 shRNA had no synergistic inhibitory effect on the growth of lung adenocarcinoma cells,while the third-line EGFR inhibitor osimertinib showed additive anti-proliferative effects.Moreover,we provided evidence that HE4 regulated EGFR expression by transcription regulation and protein interaction in LUAD.Our findings suggest that HE4 positively modulates the EGFR signaling pathway to promote growth and invasiveness in LUAD and highlight that targeting HE4 could be a novel strategy for LUAD treatment.

To editor:Fetomaternal hemorrhage(FMH)refers to the fetal blood transfection into the maternal circulation,usually involving a bleeding volume exceeding 30 ml.This phenomenon can promptly trigger a sinusoidal fetal heart rate pattern,posing a significant risk of life-threatening fetal anemia and even mortality1,2 Upon suspicion of FMH,immediate intervention is essential to prevent neonatal death,often leading to the insufficient recognition of placental examination in many published reports.However,an increasing number of FMH case reports have underscored the potential subsequent risk of choriocarcinoma.

基于TGF-β通路考察BMP4/FBN1对脊髓室管膜瘤增殖侵袭性的影响

目的基于TGF-β通路考察BMP4/FBN1对脊髓室管膜瘤(SCE)增殖侵袭性的影响及其相关机制。方法收集SCE患者肿瘤组织,同时收集正常组织作为对照,分为对照组、病例组。利用SCE患者肿瘤组织培养肿瘤细胞,并用携带FBN1 shRNA,BMP4 shRNA的慢病毒基因敲除FBN1或BMP4蛋白的表达。通过qRT-PCR和Western Blot检测肿瘤组织样本中FBN1和BMP4基因和蛋白表达水平。在BMP4低表达和FBN1低表达肿瘤细胞中通过qRT-PCR检测TGF-β基因表达水平;同时通过CCK-8细胞增殖实验与Transwell侵袭实验进一步检测敲低FBN1或BMP4蛋白的表达对肿瘤细胞增殖和侵袭能力的影响。结果与对照组相比,病例组BMP4和FBN1基因表达量升高(分别为P<0.05,P<0.01),蛋白表达量也升高(P<0.01);敲低FBN或BMP4蛋白不仅可下调SCE细胞中TGF-β基因的表达(分别为P<0.05、P<0.01),同时抑制了SCE细胞的增殖与侵袭能力(分别为P<0.05、P<0.01)。结论本研究基于TGF-β通路,考察关键分子BMP4/FBN1对SCE增殖侵袭性的影响,提示FBN1、BMP4与TGF-β之间可能存在相互作用,为临床治疗罕见病SCE提供可用的理论基础。

CT定量和定性特征联合预测肺磨玻璃结节浸润程度555例的回顾性队列研究被引量:3

目的探讨CT影像学定量和定性特征和肺磨玻璃结节浸润程度的相关性,为磨玻璃结节患者术前规划提供参考价值。方法纳入2020年9月—2022年7月期间在昆明医科大学第三附属医院行手术治疗术后确诊为肺腺癌的磨玻璃结节患者的临床资料。根据病理诊断结果将其分为两组,非浸润腺癌组(包括原位腺癌和微浸润腺癌)和浸润性腺癌组。收集其影像学特征,并进行单因素logistic回归分析,将差异有统计学意义的指标纳入多因素logistic回归分析,最终确定浸润性腺癌发生风险的独立预测因素,建立预测模型。根据约登指数计算出灵敏度和特异度。使用曲线下面积(area under the curve,AUC)值、校准曲线和决策分析曲线评价模型的区分度、校准度和临床实用价值。结果555例纳入研究。非浸润腺癌组310例,其中女235例、男75例,年龄49(43,58)岁;浸润性腺癌组245例,其中女163例、男82例,年龄53(46,61)岁。二元logistic回归分析显示,肿瘤最大径[OR=4.707,95%CI(2.060,10.758)]、实性成分占比(consolidation tumor ratio,CTR)[OR=1.027,95%CI(1.011,1.043)]、最大CT值[OR=1.025,95%CI(1.004,1.047)]、平均CT值[OR=1.035,95%CI(1.008,1.063)]、毛刺征[OR=2.055,95%CI(1.148,3.679)]、血管集束征[OR=2.508,95%CI(1.345,4.676)]是浸润性腺癌发生的独立危险因素(P<0.05)。基于上述独立预测因素构建浸润性腺癌发病风险的预测模型。模型预测公式为Logit(P)=–1.293+1.549×肿瘤最大径+0.026×CTR+0.025×最大CT值+0.034×平均CT值+0.72×毛刺征+0.919×血管集束征。模型AUC值为0.910[95%CI(0.885,0.934)],说明模型预测浸润性腺癌具有良好的区分度,校准曲线显示预测模型具有良好的校准度,决策分析曲线显示模型具有良好的临床实用性。结论联合CT定量和定性特征的预测模型对磨玻璃结节侵袭性具有良好的预测能力。其预测效能高于任意单一指标。

Global warming scenarios for the Eastern Adriatic Sea indicate a higher risk of invasiveness of non-native marine organisms relative to current climate conditions

Globally,marine bioinvasions threaten marine ecosystem structure and function,with the Mediterranean Sea being one of the most affected regions.Such invasions are expected to increase due to climate change.We conducted a risk screening of marine organisms(37 fishes,38 invertebrates,and 9 plants),both extant and‘horizon’(i.e.,not present in the area but likely to enter it).Based on expert knowledge for the Eastern Adriatic Sea coasts of Slovenia,Croatia,and Montenegro,screenings were conducted under both current and predicted climate conditions indicating with an increase in sea surface temperature and salinity of the Adriatic Sea together with changes in precipitation regime.Our aims were to:(1)identify non-native extant and horizon marine species that may pose threats to native biodiversity and(2)evaluate the risk of invasiveness of the selected species under current and predicted climate conditions.Of the 84 species screened,there was an increase in those ranked as‘high risk’from 33(39.3%)under current climate conditions and to 47(56.0%)under global warming scenarios.For those ranked as‘very high’risk,the increase was from 6(7.1%)to 21(25.0%).Amongst the screened species,the already established high-risk species Pacific oyster Magallana gigas and Atlantic blue crab Callinectes sapidus represent a threat to ecosystem services.Given the under-representation of marine species in the current European Union List,the species we have ranked as high to very high risk should be included.

儿童功能性微创理念与应用

近几十年来,微创内镜手术已经广泛应用于小儿外科各个领域,微创内镜手术具有创伤小、恢复快、痛苦少等优势,但手术切口的微创并不等于功能上的微创。本文旨在评述功能性微创的理念及其在儿童外科领域的临床应用,通过与精准医疗、新技术应用以及加速康复外科理念的结合,实现以最小创伤获得最佳治疗效果,提高患儿生活质量。

沉默PREX1表达对CHMp细胞增殖及侵袭性的影响

犬乳腺肿瘤是犬常发肿瘤之一,且有50%左右为恶性,其转移倾向明显,易引起动物死亡。Rho-GTPase蛋白家族中的PREX1蛋白,过度激活会导致Rac-GEF功能紊乱,影响细胞周期,改变细胞生理功能,PREX1的改变可能对细胞癌变有促进作用。为探讨PREX1对犬乳腺肿瘤细胞生理功能的影响,本研究使用shRNA沉默PREX1表达,利用Western blot试验检测沉默后细胞PREX1蛋白表达量的变化,通过CCK-8试验、克隆形成试验探究其对犬乳腺肿瘤细胞系CHMp增殖能力的影响,通过划痕试验和Transwell试验探索沉默其对犬乳腺肿瘤细胞系CHMp侵袭性的影响。研究结果表明,PREX1的低表达,可以抑制CHMp的增殖及侵袭能力。

影像组学联合Nomogram图预测亚实性肺结节浸润性

目的:基于CT影像组学特征联合Nomogram图构建临床诊断模型,评价其对亚实性肺结节浸润性的预测能力。方法:回顾性收集在薄层CT图像表现为肺亚实性结节并经病理证实的患者的临床及影像学资料。从CT图像中提取影像组学特征。使用LASSO回归及K折交叉验证进行特征选择。根据确定的临床独立预测因子和RS,采用多因素Logistic回归分析分别构建3种预测模型:第1种基于临床资料及影像学特征参数;第2种依据影像组学特征;第3种临床-影像组学联合模型,使用Nomogram图将Logistic回归分析的结果进行可视化表达。采用受试者工作特征曲线比较3种模型对磨玻璃样肺腺癌IA和非IA的分类预测性能。采用决策曲线分析评估不同队列中3种模型的临床实用性。结果:共收集192例共204枚亚实性肺结节,根据组织学分型分为IA 114例,非IA 90例,训练集143例(IA/非IA为77例/66例),测试集61例(IA/非IA为38例/23例)。每个亚实性肺结节可提取1316个特征,通过特征选择及Logisti回归分析最终选取2个临床独立预测因子(平均CT值、结节最大径)及3个影像组学特征用于模型构建。临床-影像组学联合模型在训练集(AUC=0.920,95%CI:0.818~0.931)中区分IA和非IA的能力优于影像组学模型和临床模型(AUC=0.907,95%CI:0.792~0.914;AUC=0.822,95%CI:0.764~0.895),测试集中临床资料的加入对提高影像组学模型的诊断效能有一定帮助。DCA表明多数情况下联合模型可以提供更大的临床效益。结论:本研究开发的临床-影像组学组学联合模型在预测亚实性肺结节的浸润性方面表现良好。

机器学习优化能谱CT预测胃腺癌的浸润性

目的探讨机器学习(ML)算法结合能谱CT定量参数和临床模型在预测胃腺癌(GAC)患者淋巴血管浸润(LVI)和神经周围浸润(PNI)的潜在价值。方法收集2017年12月—2022年5月经病理证实的GAC患者114例。研究参数涉及血清肿瘤标志物、CT-TN分期、CT评估壁外血管浸润(CT-EMVI)以及能谱CT定量参数。通过WEKA软件的Best-First算法进行特征筛选,并运用贝叶斯网络(BN)及支持向量机(SVM)算法建立模型。结果相较于LVI/PNI阴性组,LVI/PNI阳性组中CT-T_(3~4)期、CT-N阳性、CT-EMVI阳性、血清肿瘤标志物[糖类抗原(CA)72-4和CA19-9]更为常见,能谱CT参数也更高,差别均有统计学意义(P<0.05)。经特征选择,关键变量包括CT-T分期、CT-EMVI、VP-NIC和EP-70 keV CT值。基于这些变量,分别使用BN和SVM构建临床参数模型、能谱CT参数模型和混合模型,共6个模型。6个模型均展现了高预测性能,无过拟合现象。BN的混合模型预测性能最佳,AUC值为0.890~0.933,Delong检验显示其在统计学上具有显著优势(P<0.05);而SVM的混合模型与另外2种模型间的差别无统计学意义(P>0.05)。结论结合临床和能谱CT参数的ML模型能够高效能评估GAC患者的LVI和PNI状态,其中BN混合模型的准确性最高。
